虹鳟MC4R基因的PCR扩增及其应用 被引量:3

摘要 黑素细胞皮质激素受体(MC4R)是跨膜G蛋白偶联受体。MC4R在人和鼠的体重、能量稳态和采食量的调控中具有重要作用,是第一个发现的与人类显性遗传疾病性肥胖相关的靶位点。虹鳟(Oncorhynchus mykiss)属于冷水性鱼类,具有很好的药用和食用价值,但生长缓慢。本研究根据斑马鱼的MC4R基因保守区的核苷酸序列设计引物,通过PCR扩增出虹鳟的MC4R基因,纯化后测序。本实验测出虹鳟MC4R基因968bp,并发现其与其它鱼类的MC4R进行了同源性分析,构建基因进化树。 Melanocortin receptor 4 (MC4R)is one of cross membrane G protein coupling acceptor. MC4R gene was the first gene target site related human dominance heredity obese disease. It has the important function on the regulation of body weight, the energy stable state and intake the appetite in human and mouse. Rainbow trout is very valuable for medicinal purposes and as food because of its cold water environments, but it has slow growth. We act according to conservative zebra's MC4R gene sequence and design a couple of primes, clone the rainbow trout MC4R gene by PCR and sequence after purifying in this study. The 968bp sequences of the Rainbow trout MC4R gene were obtained, the homology was analyzed and evolution trees were constructed.
