摘要
目的研究维生素E琥珀酸酯(vitamin E succinate,VES)对体外培养的人晶状体上皮细胞(HLECs)增殖的影响。方法人晶状体上皮细胞株HLECs-B3以VES刺激24h、48h、72h、96h,VES浓度为5μg/ml、10μg/ml和20μg/ml,用MTT比色法测定VES对细胞增殖的抑制作用;以流式细胞仪分析细胞周期。结果VES对人晶状体上皮细胞的增殖具有显著的抑制作用,表现为时间和剂量依赖关系。并且将细胞周期阻滞于G0/G1期。结论VES对体外培养的人晶状体上皮细胞的增殖具有抑制作用,为临床筛选防治后发性白内障的药物提供依据。
Objective To investigate the proliferation inhibition effects of vitamin E suceinate (VES) on human lens epithelial cells. Methods Human lens epithelial cells were treated with VES for 24h,48h ,72h and 96h,the concentrations of VES were 5μg/ml , 10μg/ml and 20μg/ml. The inhibitory effect was measured with MTr method and the cell cell was analyzed with flow cytometry. Resets VES had significant inhibitory effect on the proliferation of human lens epithelial ceils and the effect was dependently related to time and dosage. And ceil cycle was arrested of G0/G1 phase. Conclusions VES has significantly proliferation inhibition effect on human lens epithelial cells cultured in vitro, which is helpful in treating posterior capsular opacification.
