摘要
目的观察大鼠不同时间和不同损伤程度下,损伤段脊髓内Rho—A mRNA的表达及与损伤程度的相关性。方法按WD法制模,将SD大鼠135只随机分成3组,A组为全瘫组,B组为不全瘫,C组为空白手术对照组,每组分别为45只。致伤后8h、24h、3d、7d、28d,提取损伤段脊髓内的总RNA,荧光定量PCR和凝胶电泳检测Rho—A mRNA的表达,进行大鼠后肢运动功能评分,并作相关性分析。结果(1)A、B、C组在各时间点均有Rho—A mRNA表达,但同时间点A、B组的表达明显高于C组,A组同时间点的表达高于B组,C组各时间点表达维持在较恒定水平;脊髓损伤后8hRho-A mRNA表达增加,伤后3d达高峰,持续高水平表达28d。(2)C组各时间点大鼠后肢运动功能评分明显高于A、B组,A组同时间点的评分低于B组。结论脊髓损伤后Rho—A mRNA的表达明显升高,且与脊髓损伤程度呈正相关。
Objective To observe the expression of Rho-A mRNA and correlation with the injury degree in spinal cord injury in rats. Methods 135 adult Sprague-Dawley rats randomly divided into 3 groups. 45 rats in group A were subjected to weight-drop impact causing complete paraplegia ,45 rats in group B were did with half weight causing imcomplete paraplegia and there were 45 rats in group C as sham group. The expression of Rho-A mRNA with RT-PCR, The Basso-Beattie-Bresnahan (BBB) scales of hindlimb movement were carried out respectively in three groups at 8, 24 hours and 3, 7, 28 days after spinal cord injury. The correlation about expression of Rho-A mRNA with the degree of injury were analysed. Results The expression of Rho-A mRNA was raised in group A and group B at 8 hour, reached the maximum level at 3 days and maintain high level at 28 daye after spinal cord injuries while it was observed in three groups. The level of expression of Rho-A mRNA and the BBB scales of hindlimb movement in group A was significantly higher compared to group B and it in group B was significantly higher compared to group C at 8, 24 hours and 3, 7, 28 days respectively after spinal cord injuries in rats. With regression and correlation analysis, There is significantly positive correlation between the level of expression of Rho-A mRNA and the dgree of spinal cord injuries. Conclusions Rho-A mRNA is highly expressed in spinal cord after spinal cord injuries in rats and There is significantly positive correlation between it's level and the dgree of spinal cord injuries.
