摘要
Rho通过GTP结合形式和GDP结合形式的转换起着分子开关的作用,调控许多细胞内信号通路。Rho通过激活其下游靶分子Rho激酶调节细胞的收缩、黏附、迁移、增殖、凋亡等多种生物学行为和功能。目前已证实Rho/Rho激酶通路参与肺动脉高压、哮喘、特发性肺纤维化和肺癌等多种肺部疾病的病理生理过程,在这些疾病的动物模型中阻断该信号通路能够改善这些疾病的病理改变与预后。
Rho exhibits both GDP/GTP-binding activity and functions as a molecular switch, cycling between a GDP-bound inactive state and a GTP-bound active state. Activated RhoA via its downstream effector Rho-kinase is involved in a wide range of cellular functions, such as cell contraction, adhesion, migration, proliferation and apoptosis. Recently, accumulating evidence has been suggested that activation of the Rho/Rho-kinase signaling pathway is substantially involved in the pathogenesis of several pulmonary diseases, such as pulmonary hypertension, asthma, idiopathic pulmonary fibrosis and lung cancer, while inhibitors of Rho/Rho-kinase signaling pathway can improve the outcome of these diseases in animal models.
