摘要
目的探讨TGF-β1在肾组织纤维化中的作用机制。方法采用原代培养的人肾成纤维细胞,将细胞分为0、0.1、1、5ng/ml和10ng/ml不同浓度TGF-β1刺激培养组。应用免疫细胞化学方法检测TGF-β1作用下人肾成纤维细胞热休克蛋白47(heat shock protein 47,HSP47)表达,RT-PCR方法检测TGF-β1作用下人肾成纤维细胞HSP47和Ⅰ型胶原mRNA表达。结果(1)对照组(0ng/ml TGF-β1组)人肾成纤维细胞几乎无HSP47表达,在TGF-β1刺激下,培养的人肾成纤维细胞HSP47表达增强。(2)在0.1~5ng/ml范围内,TGF-β1以剂量依赖方式促进培养的人肾间质成纤维细胞表达HSP47和Ⅰ型胶原mRNA。结论TGF-β1促进肾组织纤维化形成,可能部分是通过其促进肾间质成纤维细胞表达HSP47,进而增加胶原合成所致。
Objective To investigate the mechanism of TGF-β1 action in chronic renal fibrosis. Methods Human renal fibroblasts primarily cultured in vitro were divided into 5 groups: (1) the control group with 0ng/ml TGF-β1 ; (2) 0. lgng/ml TGF-β1 trial group; (3)lng/ml TGF-β1 trial group; (4)5ng/ml TGF-β1 trial group; (5)10ng/ml TGF-β1 trial group. Expression of heat shock protein 47(HSP47), which in human renal fibroblasts in vitro under the effect of TGF-β1 , was determined by immunocytochemistry. And expressions of HSP47 mRNAs and I collagen mRNAs were examined by semi-quantitive reverse transcriptase polymerase chain reaction (RT- PCR). Results Compared with the control group, expression of HSP47 and collagen Ⅰ in cultured human renal fibroblasts significantly increased in the trial groups. TGF-β1 promoted expression of collagen I and HSP47 of renal fibroblast in dose-dependent manner range from 0.1ng/ml to 5ng/ml. Conclusion TGF-β1 is involved in occurrence and development of renal fibrosis partly by enhancing expression of HSP47 in renal fibroblasts and further increasing collagen synthesis.
