摘要
目的建立反相高效液相色谱法测定妥布霉素滴眼液含量的方法。方法选用C18色谱柱,2,4-二硝基氟苯(DNFB)柱前衍生化,0.25%三羟甲氨基甲烷-乙腈-0.5mol·L^-1硫酸溶液(40:59:1)为流动相,检测波长为365nm。结果在10.56-15.84μg范围内峰面积与浓度呈良好线性关系,r=0.9996;平均回收率为99,90%。RSD为0.37%。结论该方法简便准确,重现性好,可用于妥布霉素滴眼液含量的测定。
Objective To determine the content of tobramycin in eye drops by RP-HPLC. Methods The precolumn derivation using 2,4-dinitroflurobenzene(DNFB) was carried out. The chromatographic separation was performed in C18 column. The mobile phase was 0, 25% tris(hydroxymethyl) aminomethane-ancetonitrile-0. 5 mol·L^-1 sulfuric acid(40: 59: 1 ). The detection wave length was 365 nm. Results The linear range was 10. 56 - 15. 84 μg ( r = 0. 999 6). The average recovery was 99.90% (RSD = 0. 37% ). Conclusion The method was simpl, accurate and highly reproducible , and can be used for the quality control of tobramycin in eye drops.
