摘要
本试验发现10μg/ml阿霉素可诱导人粘液表皮样癌MEC-1细胞凋亡,在用流式细胞仪(FCM)测细胞DNA变化及p53蛋白表达时,p53蛋白表达和凋亡细胞随着药物作用时间的延长而剧增,p53的免疫组化也说明了阿霉素可使细胞中p53蛋白表达增强。这些结果揭示了阿霉素诱导的MEC-1细胞凋亡为p53依赖性的。p53蛋白与细胞凋亡之间关系密切,也为基因疗法和凋亡的机理提供了一定的理论依据。
The induction of apoptosis and expression of p53 protein in mucoepidermoid carcinomaMEC-1 cell line by Doxorubicin was studied. The flow cytometric assay was used in thisstudy. After 24h treatment with Doxorubicin, the percentage of apoptosis and the expressionof p53 protein were increased significantly. The data demonstrated that Doxorubicin inducedapoptosis of MEC-1 cell was p53 dependent, and there was a close relationship between p53and apoptosis.
