摘要
目的探讨葛根素对大鼠视网膜缺血再灌注损伤中肿瘤坏死因子-α(TNF-α)表达的影响及对视网膜的保护作用。方法建立大鼠视网膜缺血再灌注模型,分缺血再灌注组、缺血再灌注+葛根素组和对照组。每组按再灌注时间的不同分为缺血再灌注1h、6h、12h、24h、48h、72h组。以免疫组织化学法检测各期视网膜中TNF-α的表达,同时记录各期大鼠ERGa、b波波幅。结果在缺血再灌注组,TNF-α在再灌注6h后开始表达,24h表达最强,以后逐渐下降。在缺血再灌注+葛根素组,在再灌注6h可见TNF-α微弱表达,再灌注12h后开始表达较为明显,24h表达最强,但明显低于缺血再灌注未处理组同期TNF-α的表达。对照组中未见明显的TNF-α的表达。同时,缺血再灌注+葛根素组各时期ERGa、b波波幅高于缺血再灌注组而低于对照组。结论葛根素对大鼠视网膜缺血再灌注损伤中TNF-α的表达具有抑制作用。
[Objective] To study the effect of puerarin on the expression of TNF-α in rat's ischemia-reperfusion injuried retina and its protection for rat's retina. [Methodsl The model of retinal ischemia-reperfusion was emploied by ligationed the left side of artery, the rats were devided into isehemia-reperfusion group, ischemia-reperfusion and puerarin group and control group, each group including 1 h, 6 h, 12 h, 24 h, 48h and 72 h. Immunohisto-chemistry was used to detect the expression of TNF-α protein in different stages of rat's retina. The ERG were examined in different stages. [Result] In ischemia-reperfusion group, TNF-α began to express after reperfusion for 6 h and increased gradually, it reached the highest level after reperfusion for 24 h. In ischemia-reperfusion and puerarin group, TNF-α can be detected at 12 h, its expression strongest at 24 h. But was lower than the same stage of ischemia-reperfusion group. There was no TNF-α expressed in control group. The relative ERG a, b wave of isehemia-reperfusion and puerarin group was higher than ischemia-reperfusion group, but lower than the control group. [Conclusion] The puerarin can inhibit the expression of TNF-α in rat's ischemia-reperfusion injuried retina and protect it.
