摘要
应用直流电电解法直接破坏大鼠下丘脑腹内侧核(VMH)复制肥胖模型。VMH损伤后立即进行静脉葡萄糖耐量试验(ivGTT),观察到葡萄糖注射后10min,胰岛素分泌即明显升高,到15min时,胰岛素水平高于对照组,但无差别。血浆葡萄糖水平在ivGTT试验的各时点均无差别。VMH损伤后1天即出现高胰岛素血症,此后仍不断上升。基础胰岛素水平升高与摄食量增加有明显的相关关系(r=0.6756,P<0.01).VMH损伤后第1天和第5天分别出现高甘油三酯血症、高胆固醇血症。
n obesity model was used by eletrolytical destruction of bilateral ven-tromedial hypothalamus(VMIi)in ruts.VMH obesity rats have characte-ristics of hypcrphagia,easy anxiousnes,hyperinsulinemia and abnormal lip-id metabolism etc.Net body weight inereased 40g within the first week after VMH lesions.Just after VMH lesion,an iv glucose tolerance test was carr-ied out,and 10 minutes later,insulin secretion elevated significantly,and by 15 minutes,insulin level was slightly higher than that of the control group(P>0.05).Meanwhile plasma glucose leveis were appromixately simi-lar between the twogroups at any time points.Hyperinsulinelnia occurred 1 day postlesions,and kept increasing trend during the subsequent several days.Therc was a significant correlation between the increase of insulin le-vel and food intake after VMH lesion. Hypertriglyceridemia and hypercho-lesterolemia developed on the first and fifth day after VMH lesions respectively.
