摘要
目的建立人前列腺癌裸鼠骨转移动物模型。方法采用人前列腺癌细胞系PC-3细胞悬液,分别通过胫骨髓腔注射、左心室注射及尾静脉注射3种方法接种于裸鼠体内,定期进行影像学观测;8~16周后处死动物行病理切片检查,观察骨转移情况。结果经影像学检查和病理组织学鉴定,PC-3细胞经胫骨髓腔注射后,该组裸鼠于第2周出现骨转移表现,转移率为100%(7/7);左心室注射后,至第12周影像学检测到骨转移率为28.6%(2/7),处死动物后经病理学检查骨转移率为42.9%(3/7);而尾静脉注射组影像学和组织病理学检查均未见骨转移(0/7)。结论胫骨髓腔注射和左心室注射两种方法均能在短期内建立人前列腺癌骨转移动物模型。
Objective To establish animal bone metastasis models of human prostate cancer. Methods Human prostate cancer cell line PC-3 cells were injected into the bone marrow cavity of the right tibia, the left cardiac ventricle or the tail vein of BALB/C-nu/nu nude mice, respectively and the mice were observed radiographically every week. After 8 to 16 weeks, the animals were killed and pathological sections of bone tissues were made for observation of bone metastasis. Results After PC- 3 cells were injected into the bone marrow cavity of the tibias, radiographic and pathohistological examinations revealed that bone metastasis was detected in the right tibias of all nude mice (7/7 mice) at second week; Bone metastasis in 2 nude mice (2/ 7 mice) was detected by X-ray at 12th week after PC-3 cells were injected into the left cardiac ventricle of nude mice; And no bone metastasis was observed at 16th week after PC-3 cells were injected into the tail vein of nude mice. All bone metastases were confirmed by HE staining and immunohistochemical staining of anti-cytokeratin 8/18. Conclusion The animal bone metastasis models of human prostate cancer could be established in a short period by injecting PC-3 cells directly into the bone marrow cavity of tibia or left cardiac ventricle of nude mice.
