山羊卵子发育、受精和老化过程的电镜组化研究 被引量:1

ELECTRON MICROSCOPIC HISTOCHEMISTRY OF THE GOAT OOCYTE DURING DEVELOPMENT, FERTILIZATION AND AGING

摘要 应用改良柠檬酸铅法、Tris-Malcate法和Dermer氏保存磷脂的固定方法,分别研究了山羊卵母细胞发育、受精和老化过程中硷性磷酸酶(Alpase)、酸性磷酸酶(Acpase)和磷脂的超微结构定位和含量。结果表明,a.碱性磷酸酶主要定位于MV.卵皮质和伸入到卵母细胞内的颗粒细胞突起上;生长卵母细胞的硷性酸酶活性很强,排卵后活性减弱,受精卵中无Alpase活性。b.排卵24小时后卵母细胞内开始出现Acpase阳性的髓样小体,并随卵老化而逐渐增多。c.卵母细胞脂滴内磷脂含量很少,且恒定;MV、ZP、质膜和Mi上都含有许多磷脂;生长卵母细胞MV和质膜上磷脂多,排卵后减少;受精卵质膜和Mi膜上磷脂明显增多。文中讨论了酶和磷脂变化与卵母细胞生理状态的关系。 <ABSTRACT> The ultrasturctural localization and activity of alkaline phosphatasc (Alpasc),acidphosphatasc (Acpase) and Phospholipids in thc goat oocytc during its dcvclopment, l^crtiliza-tion and aging were studied by means o^thc Modified Lead Citratc method, thc T.ris-Malaetcmethod and Dcrmcr^s fixation for phospholipids, respectively. The results indicated that: (1)Thc Alpasc activity was mainly localizcd on the mierovilli and cotical cytoplasm oF the oocytcand on thc follicular ecl1 processes which extended ~to thc oocytc. Thc Alpasc was highly ac-tive in the developing ooeytc and thc activity was decreasing alter ovulation. No Alpase activ-ity was dctccted in thc fertilized egg; (2) Thc myelin bodies with Aepasc activity began to ap-pear in thc oocytes 24 hours a^tcr ovulation and the number increased as the oocytes becamemore aged; (3) Thc lipid drops in thc goat oocytc contained a mild amount or phosphlipidsand thc amount rcmaincd unchangcd during the oocytc development, ~ertilization and aging.Considerable amount of phospholipids was also found in thc microvilli, zona pellueida, plasmmcmbranc and thc mitochondria. The phospholipids on the MV and thc plasma mcmbrancwcrc thc most obvious and thc amount dcelincd aftcr ovulation. After fcrtilization, theamount of phospolipids on thc cell mcmbranc and thc mitocondria incrcascd remarkably. At-tempts wcrc madc in thc discussion to correlate thc changcs in thc enzymes and phospholipidsduring oocyte devclopmcnt, fcrtilization and aging with the physiological states of thc oocyte.
