Tca8113和BcaCD885对口腔癌靶向隐形顺铂聚乳酸纳米粒的敏感性研究 被引量:1

摘要 目的:观察口腔鳞癌细胞系(Tca8113和BcaCD885)对口腔癌靶向顺铂聚乳酸聚乙二醇纳米粒(CDDP-PLA-PEG-NP,或称隐形聚乳酸纳米)的敏感性,并与顺铂(CDDP)对Tca8113和BcaCD885的抗癌活性进行对比观察。方法:用四唑盐显色法(MTT法)检测CDDP-PLA-PEG-NP和CDDP在1-8 d内8个时间点对Tca8113和BcaCD885的抗癌活性,计算出2种药物在8个时间点对2种癌细胞的抑癌率和药物的半数抑癌率浓度。结果:CDDP-PLA-PEG-NP和CDDP对Tca8113和BcaCD885均有强的杀伤作用,其杀伤效应均呈时间依赖性和剂量依赖性。结论:经改型后的CDDP-PLA-PEG-NP对口腔鳞癌细胞Tca8113和BcaCD885仍有强大的杀伤作用,为以后进一步的体内实验提供了依据。 Objective: To study chemosensitivity and anticancer activity of Cisplatin (CDDP) loaded polylactic acid - polyethylene glycol nanoparticles (CDDP- PLA- PEG- NP) against oral squamous carcinoma cell lines Tca8113 (tongue carcinoma) and BcaCD885 (buccal carcinoma). Methods: The chemosensitivity of Tca8113 and BcaCD885 to CDDP- PLA -PEG- NP and CDDP was tested using MTT assay. The suppressed cancer rate and 50% inhibition concentrations values (IC50) of two drugs against two cancer cell lines were counted for 1,2,3,4,5,6,7and 8 days at different concentrations. Results: Both CDDP -PLA -PEG -NP and CDDP showed high anticancer effects against Tca8113 and BcaCD855, the anticancer activity of two drugs was both dose and time dependent. Conclusion: CDDP within CDDP - PLA - PEG - NP remained its anticancer effects. And CDDP - PLA - PEG - NP showed higher than that of CDDP.
