摘要
目的:进一步探讨糖皮质激素防治肾间质纤维化的机理。方法:在成功分离培养人肾间质成纤维细胞基础上,通过流式细胞仪分析、dUTP标记DNA断裂点显色及电镜技术等方法,观察甲基强的松龙对肾间质成纤维细胞编程性死亡的影响。结果:甲基强的松龙处理组编程性死亡的细胞百分数明显高于对照组,且呈时间依赖性。结论:甲基强的松龙可促进人肾间质成纤维细胞的凋亡。
BJECTIVESRecently,ithasbenshownthatabnormalgrowthandapoptosisoffibroblastsplayapivotalroleinrenalinterstitialfibrogenesis.Withtheestablishmentofhumanrenalinterstitialfibrob-lastscultures,weobservedtheefectsofglucocorticoidsontheprogrammedceldeathofthesehumanfibroblasts.METHODOLOGYHumanrenalinterstitialfibroblastsderivedfromkidneyswithinterstitialfibrosiswereculturedrandomlywithorwithout10-3Mmethylprednisolone.Programmedceldeathwasdetect-edwithflowcytometricDNAanalysis,dUTPlabelingDNAbreakingpointsandelectronicmicroscopicobservation.RESULTSMethylprednisolonesignificantlypromotedtheapoptosisofrenalinterstitialfibroblastsinvitroinatime-dependentmannerascomparedtothecontrol.Apoptoticcelsdetectedbyflowcytome-trywere1.8%and25.8%oftotalcelsat12hours;2.3%and68.9%at24hoursforthecontrolgroupandmethylprednisolonetreatedgrouprespectively.CONCLUSIONSItwasindicatedthatinducingapoptosisoffibroblastsmaybeanimportantmecha-nismformethylprednisolonetopreventrenalinterstitialfibrosis.
