摘要
用选择性系列抽提技术与整装制样方法相结合,清晰地显示了Hela细胞与BHK_(-21)细胞的核骨架—Lamina—中间纤维结构体系。在此基础上,用电镜免疫胶体金标记技术对这一精细结构体系的成份分别进行定性研究,实验结果说明:Hela细胞与BHK_(-21)细胞核骨架纤维均能被298KD的核骨架蛋白单抗—金颗粒标记,它们的Lamina均被LaminA单抗—金颗粒标记,Hela细胞的中间纤维既能被角蛋白单抗又能被波形蛋白单抗—金颗粒所标记,而BHk_(-21)细胞的中间纤维却只能被波形蛋白单抗—金颗粒标记,不能被角蛋白单抗—金颗粒标记,但BHK_(-21)细胞的Lamina却与角蛋白单抗有交叉反应。将核骨架—Lamina—中间纤维体系的精细结构与其蛋白成份的定性同时准确地显示出来,至今尚未见过报导。
Using serial extraction technique and whole mount cell technique, we clearly show the nuclear matrix-Lamina-intermediate filament system of Hela cell and BHK_2~cell. Based on this, we apply immuno-gold electron matrix filaments of both Hela cell and BHK_2~cell can be labelled with monoclonal anti-298KD nuclear matrix protein-gold particles; their Lamina can all be labelled with monoclonal anti-lamin A-gold particles; the inter-mediate filaments of Hela cell can be labelled with monoclonal anti-Keratin and anti-Vimentin-gold particles, but the inter-mediate filaments of BHK_zt cell can only be labelled with anti-Vimentin not anti-Keratin monoclonal antibody-gold-particles. Nevertheless, the Lamina of BHK_2~ cell can react crossly with monoclonal anti-Keratin. Until now, there is no report about the showing simultaneously of both the fine structure of nuclear matrix-Lamina-intermediate filament sys- tem and chracterization of its protein composition exactly.
