葡多酚对体外培养肝细胞PKC表达影响

摘要 目的体外观察葡多酚(GPC)对小鼠肝细胞蛋白激酶C(PKC)表达的影响。方法将正常小鼠肝细胞加入不同剂量GPC共同培养后,用免疫细胞化学法检测各组肝细胞PKC表达;四甲基偶氮噻唑蓝(MTT)法检测各组肝细胞的增殖活性。结果100 mg/L GPC组肝细胞内PKC阳性表达率和细胞增殖活性分别为34.24%和0.654±0.062,经统计学χ2检验和t检验,与正常对照比较差异均有统计学意义(P<0.05)。结论葡多酚可促进小鼠肝细胞PKC表达,提高肝细胞增殖活性。 Objective To study the effect of grape procyanidins(GPC)on the protein kinase C (PKC) expression in mice' s hepatic cells in viro. Methods The nomal mice's hepatic cells were cultured with different dose of GPC. The proliferation activity of the hepatic cells was determined by MTT assay, and the levels of PKC protein expression in the cell were measured by immunoeytochemistry method. Results The expression rates of PKC protein and the proliferative activity of 100 mg/L GPC group were 34.24 % and 0. 654 ± 0. 062, which were significantly higher than those of the control group by x^2 teat and t test (P 〈 0.05). Conclusion GPC can promote the PKC protein expression and improve the proliferative activity of the mice's hepatic cells.
