摘要
目的研究慢性脑缺血Orexin受体-2(OX2R)的表达。方法通过结扎双侧颈总动脉建立脑缺血模型;用水迷宫进行行为学评价;双标免疫荧光观察OX2R的表达。结果缺血15d时大鼠的学习记忆能力明显减退,1个月、2个月较缺血15d模型组学习记忆能力有所好转。同时,缺血15d时OX2R在神经元的表达明显增高,1个月时OX2R的表达较15d时明显下降,2个月时OX2R的表达再次增高;此外,OX2R在神经纤维亦有表达其变化趋势同神经元。组织学示15d时部分细胞萎缩,1个月时大部分细胞变形萎缩,2月时部分细胞形态恢复正常。结论提示脑缺血的早期OX2R可能有诱导细胞凋亡的作用,而缺血的后期有神经保护作用。
Objective To study the OX2R expression of chronic cerebral ischemia. Methods Established the cerebral ischemia model through ligating double carotid arteries,evaluated the intelligence by morrize maze,and observed the OX2 R expression by double immunofluorescent staining. Results The intelligence of 15d model impaired obviously, 1m and 2m improved compared with 15d model. At the same time, the OX2R expression of 15d model increased significantly , 1m model decreased obviously compared with 15d model,2m model increased obviously the second time. Except that ,neurofibra also had OX2R expression and the tendency of change was same as the neuron expression. From the histology,partial neurons of 15d model got atrophy, most neurons of 1m model. got eytomorphosis and atrophy,however partial neurons of 2m model recovered normal. Conclusion It suggests that OX2R maybe induce the apoptosis at the early stage of cerebral ischemia,however OX2R may have neuron protection at the late stage of ischemia.
