文拉法新对慢性应激引起海马CA3区细胞萎缩与凋亡的干预效应 被引量:6

摘要 目的:研究文拉法新对慢性应激引起的海马CA3区萎缩和凋亡的预防与治疗作用。方法:实验于2004-06/2004-10在中南大学湘雅医学院病理学实验室完成,将SD大鼠随机分成对照组和3个实验组,用强迫性游泳4周的方法对大鼠进行慢性应激处理,断头处死后用石蜡切片苏木精-伊红染色法对海马CA3区组织进行切片染色,在电镜下观察CA3区神经元情况。同时观察各组实验期间的行为。结果:生理盐水对照组大鼠行为均表现正常;应激+生理盐水组大鼠均出现紧张度增加,探究行为受抑制,兴趣丧失或怪异行为;应激+阿米替林组多数出现探究行为抑制、兴趣减少等情况,未见怪异行为;应激+文拉法新组少数出现行为抑制,多数大鼠探究行为正常,未见震颤或怪异行为。生理盐水对照组未出现凋亡,其他各组发生凋亡的情况分别为:应激+生理盐水组:10/16(62%),应激+阿米替林组:6/16(38%),应激+文拉法新组:3/16(19%);经统计学检验表明,应激+阿米替林组出现凋亡的大鼠数和应激+生理盐水组差异没有显著性意义(P>0.05),和应激+文拉法新组差异也没有显著性意义(P>0.05),应激+文拉法新组和应激+生理盐水组差异存在显著性意义(χ2=6.652,P<0.05)。结论:文拉法新对于慢性应激所致的CA3区细胞萎缩与凋亡具有预防和治疗作用,且对探究行为无明显影响。 AIM:To study the prophylactic and therapeutic effects of venlafaxine on chroni c stress induced cell shrinkage and apoptosis in hippocampal CA3 region. METHODS:The experiment was done in the pathological laboratory of Xiangya Scho ol of Medicine,Central South University between June and October 2004.SD rats we re randomly grouped into control group and three experimental groups,the experim ental groups were stressed by compulsory swimming every day for 4 weeks,after de capitation to all the rats,the tissue in hippocampal CA3 region of rats was sect ion stained by paraffin section hematoxylin and eosin(HE) staining methods.The status of neurons in the CA3 region was observed under electron microscope.At th e same time,the behaviors of rats were observed during the experiment. RESULTS:Rats in the saline control group showed normal behaviors; Rats in the stress+saline group showed nervous,behaviors of probing inhibition, interests l oss or unusual behaviors;Rats in the stress+amitriptyline group showed inhibiti on of probing behaviors,interests loss,but no abnormal behaviors;A few rats in t he stress+venlafaxine group showed behavioral inhibition,most rats showed norma l probing behaviors,and no tremor or abnormal behaviors.The rate of shrinkage an d apoptosis in the saline control group was zero,stress+saline group was 10/16( 62%), stress+amitriptyline group was 6/16(38%),stress+venlafaxine group was 3/16(19%).The results of statistical analysis showed that there was no signific ant difference between the stress+amitriptyline group and stress+saline group in the number of rats with shrinkage and apoptosis(P >0.05), also between the st ress+amitriptyline group and stress+venlafaxine group(P >0.05),while significa nt difference was observed between the stress+venlafaxine group and stress+sal ine group(χ2=6.652,P< 0.05). CONCLUSION:Venlafaxine has therapeutic and prophylactic effects on cell shrink age and apoptosis caused by chronic stress in hippocampal CA3 region,and do not affect its probing behaviors.
