摘要
目的探讨花边莲提取液对人肺癌SPC-A-1细胞凋亡相关基因survivin和caspase-3表达的影响。方法采用体外培养人肺癌SPC-A-1细胞,实验随机分为正常对照组、花边莲处理组(浓度分别为12.5,25和50 m l/L)和顺铂(DDp25mg/L)组,半定量RT-PCR检测survivin和caspase-3 mRNA表达水平,二步法免疫组化检测Survivin和Caspase-3蛋白表达变化。结果与正常对照组相比,花边莲各浓度组caspase-3 mRNA和蛋白表达显著上升(P<0.01),survivin mRNA和蛋白表达明显下降(P<0.01)。结论花边莲提取液诱导SPC-A-1细胞凋亡的分子机理可能与上调caspase-3基因表达和下调survivin基因表达有关。
Objective To investigate the influence of HuaBianLian(HBL) extract on expression of apoptosis associated genes survivin and caspase-3 in human lung cancer SPC-A-1 cells. Methods Cultured human lung cancer SPC-A-1 cells were randomly divided into the control group ( the negative control group), HBL - treated groups( 12.5,2.5,50 ml/L) and DDP(25 mg/L) group (the positive control group). The expression of survivin and caspase-3 mRNA was detected by semi-quantitive RT-PCR . The expression of survivin and caspase - 3 proteins were detected by two-step immunhistochemical staining. Results Compared with control group , the expression of caspase -3 mRNA and protein increased significanty ( P 〈 0.01 ), while survivin mRNA and protein decreased markedly in HBL groups ( P 〈 0.01 ). Conclusion The molecular mechanism of HBL apoptosis -inducing of SPC-A-1 cells may be associated with the up-regulating expression of caspase - 3 gene and down-regulating expression of survivin gene.
