兔视网膜中蛋白激酶C的定位研究 被引量:1

摘要 目的:证实兔视网膜中有α、β和γ三种蛋白激酶C(proteinkinaseC,PKC)亚型的分布。方法:采用免疫组织化学技术,以抗PKC同功酶Ⅰ(α)、Ⅱ(β)、Ⅲ(γ)单克隆抗体对兔视网膜进行PKC同功酶定位研究。结果:兔视网膜中有PKC-α、PKC-β和PKC-γ的阳性免疫反应。PKC-α免疫反应主要呈现在内核层的双极细胞;PKC-β免疫反应主要呈现在神经节细胞层的神经节细胞;PKC-γ则在神经节细胞层、内网状层以及光感受器的外节呈弥漫性弱阳性染色。结论:作为中枢神经系统的一部分,视网膜可以同时存在PKC-γ、PKC-β和PKC-α三种亚型。 PURPOSE:Toverifyexistanceofα-,β-,andγ-proteinkinaseC(PKC)subspeciesandtheirlocalizationinrabbitretina.METHODS:Usinganimmunohistochemicaltechniquewithmono-clonalantibodiesagainstPKCisozymes-Ⅰ(α),-Ⅱ(β),and-Ⅲ(γ)tocharacterizethedistributionofPKCinrabbitretina.RESULTS:Thereisapositiveimmunostainingforα-,β-,andγ-PKCinrabbitretina.Theimmunoreactivityofα-PKCwasobservedmainlyinthebipolarcelsofinnernuclearlay-erandtheoutersegmentsofphotorecptors.Thepositiveimmunostainingofβ-PKCcouldbeseenintheganglioncels,innerplexiformlayer,innernuclearlayer,andtheoutersegmentsofphotorecep-tors.Adifuseandweakstainingofγ-PKCisrecognizedintheganglioncellayer,innerplexifromlayer,innernuclearlayer,andtheoutersegmentsofphotoreceptors.CONCLUSION:Theproteinki-naseCsubspecies-α,-β,and-γarepresentinretinawhichisapartofthecentralnervoussystem.
