摘要 目的研究大蒜提取物二烯丙基二硫化物(DADS)诱导人非小细胞性肺癌细胞H1299凋亡及丝裂原活化蛋白激酶(MAPK)对此过程的影响,探讨DADS抗肿瘤的可能机制。方法用MTT法检测DADS处理24 h后H1299细胞活性;用蛋白质印迹法测定H1299细胞内磷酸化-p42/44 MAPK的表达。结果与对照组相比,DADS呈浓度依赖性诱导H1299细胞凋亡;蛋白质印迹法分析显示,DADS可激活p42/44 MAPK,且磷酸化-p42/44 MAPK的表达呈浓度依赖性增加。结论DADS能诱导H1299人非小细胞性肺癌细胞发生细胞凋亡,并呈浓度依赖性诱导磷酸化-p42/44 MAPK表达。 Objective To explore the antineoplastic mechanism of diallyl disulfide (DADS) through studying the apoptosis induced by DADS extracted from garlic in human lung cancer H1299 cells', Methods Cytoactive was determined with MTT method after a 24 h treatment of H1299 by DADS. The expression of phospho-p42/44 MAPK of H1299 cell was measured by western blot. Results Compared with control group, DADS could induce the apoptosis and the expression of phospho-p42/44 MAPK in human H1299 cells in a concentration-dependent manner. Conclusion DADS can induce apoptosis and the expression of Dhospho-o42/44 MAPK in human H1299 cells in a concentration-dependent manner.
