采用ADVIA 120血球分析仪及流式细胞仪评估枸椽酸钠、EDTA及CTAD三种抗凝剂对血小板激活的影响 被引量:5

摘要 目的对比枸橼酸钠、EDTA及CTAD三种抗凝剂在押制血小板体外激活的作用以覆探讨ADVIA 120血球分析仪MPC参数用于观察血小板激活的作用。方法采用ADVIA 120血球分析仪MPC参数与流式血小板CD62P检测,观察枸橼酸钠、EDTA及CTAD三种抗凝剂抗凝的86名体检者静脉血样中血小板对凝血酶的诱导激活以及自发激活作用。结果加入凝血酶后。用CTAD抗凝的全血血小板CD62表达率及MPC无显著性改变,枸橼酸钠和EDTA抗凝血均出现明显改变。时分离自CTAD抗凝血的血小板进行凝血酶激活实验,结果发现血小板能够被激活,表明CTAD抗凝血的血小板具有生物学功能。此外,分离自EDTA抗凝全血的血小板的活化标志CD62表达率在加凝血酶前明显高于CTAD抗凝全血的血小板.说明CTAD能够保护血小板免于由于血小板分离操作所致的物理激活。血小板自发激活实验中CTAD能够在体外〈Bh抑制血小板自发激活,而枸橼酸钠和EDTA抗凝血只能保证1~2h内无血小板自发澈活。ADVIA 120 MPC值的变化与流式血小板活化标志CD62检测结果存在着良好的相关性。结论CTAD抗凝剂在能够保证血小板活性的情况下抑制血小板体外生物和物理激活,是一种易于在临床推扩的用于血小板分析的抗凝剂。ADVIA 120的MPC参数检测可以用于血小板活化观察。
