摘要
目的:观察顽疣净含药血清对对郎格汉斯细胞(LC)刺激T细胞增殖和分泌Th1/Th2细胞因子的影响。方法:以顽疣净含药血清干预分离的人表皮LC。用同种异体混合T淋巴细胞反应,观察顽疣净含药血清对LC刺激T淋巴细胞增殖的影响;用ELISA法检测混合T淋巴细胞反应上清中IFN-γ和IL-10表达,了解顽疣净含药血清对LC调节Th分化的影响。结果:顽疣净含药血清干预后的LC刺激T细胞增殖能力提高,能提高混合T淋巴细胞反应上清中IFN-γ表达,抑制IL-10分泌。结论:顽疣净含药血清体外具有调整LC功能的作用。
Objective： To observe the effect of a traditional Chinese medicine, Wanyoujing, medicated serum on Langerhans＇ cells （LCs） stimulating T lymphocytes proliferation and the secretion of Th1/Th2 cytokines. Methods： With the method of serum pharmaceutics, the Wanyoujing medicated serum was used to treat the separated LCs. Mixed T lymphocytes reaction （MLR） was used to detect T lymphocytes proliferation induced by Wanyoujing medicated serum. ELISA method was used to detect the levels of IFN-γ, and IL-10 in the supernate of MLR in order to observe the effect of Wanyoujing medicated serum on Th1/Th2 cytokines. Results： The stimulating activity of LCs on the T lymphocytes proliferation increased after treated by Wanyoujing medicated serum. Through detecting IL-10 and IFN-γ, in the supernate of reaction, it was found that Wanyoujing medicated serum could enhance the secretion of IFN-γ, and inhibit the secretion of IL-10. Conclusion. Wanyoujing is effective in regulating LC＇s function in vitro.
