摘要
目的:建立反高效液相色谱法测定花锚中落干酸和龙胆苦苷的含量。方法:采用kromasil—C18(250mm×4.6mm,5μm)色谱柱,以流动相甲醇和水(含0.04%磷酸)的比例在0~15min内由23:77至30:70线性梯度洗脱,流速1ml/min,检测波长238nm,柱温35℃。结果:两种成分均达到基线分离,落干酸和龙胆苦苷的线性范围分别为0.198~5.544μg(r=0.9999),0.0262~0.7336μg(r=0.9999);回收率为96.12%(RSD=1.73%)、97.67%(RSD=1.91%)。结论:方法测定快速,结果准确、可靠。
Objective: To establish a quantitative method of simultaneously determination of loganic acid and gentiopicroside by RP-HPLC. Method :The sample wee separated on the column of kromasil-C18 (250mm × 4,6mm, 5μm)which eluted with methanol and water (0.04 % phosphoric acid), The ratio of methanol and water increased from 23:77 to 30:70 in 15 min with detected wavelength at 238nm, flow rate at 1 ml/min, column temperature at 350C. Results:Two compounds were baseisolated. The linear ranges of loganic acid, gentiopicroside were 0.198- 5. 544 μg( r= 0.9999), 0.0262= 0,7336μg( r = 0.9999) ; The average recoveries were 96.12 % (RSD = 1.73 % ) ,97.67 % (RSD = 1.91% ). Conclusions: The method is rapid and precise.
