摘要
[目的]探讨大鼠嗅神经鞘细胞条件培养液对同种大鼠骨髓基质干细胞的诱导分化作用。[方法]通过差速贴壁法获得较高纯度的嗅鞘细胞,在接种培养后的9~12d,收集并制备嗅鞘细胞条件培养液,与纯化培养第3代的骨髓基质干细胞共培养,观察其诱导分化后的形态学变化,免疫荧光染色鉴定诱导分化结果。[结果]发现大鼠嗅鞘细胞条件培养液对同种大鼠骨髓基质干细胞有明显的诱导分化作用,在72h时荧光显微镜下观察并计算MAP-2和GFAP阳性细胞的比率分别为55%、23%。[结论]大鼠嗅鞘细胞条件培养液有明显的诱导同种大鼠骨髓基质干细胞向神经样细胞分化的作用。
[ Objective] To explore the differentiation of bone marrow stromal cells (BMSCs) induced by OECs conditioned medium. [ Method] The purified OECs were acquired by seleclive culture medium and cultivated in the polylysine coated plate. After 9 -12 days culture, OECs conditioned medium was obtained by 2000 r/min configuration and then co-cultivated with purified BM SCs which were 3th generation. The differentiation of BMSCs were observed under inverted microscope and were identified by immu- mofluorescence method. [ Result] The OECs conditioned medium had obviously effects on BMSCs'differentiation . BMSCs differentiation into neuron-like cells ratio was 55% and differentiation into astroctes cells ratio was 23%. [ Conclusion] The conditioned medium which comes from OECs culture supernatant has obviously effects on BMSCs'differentiation into neuron-like cells.
