摘要
目的探讨应用四氢叶酸(CF)解救大剂量甲氨喋呤(HD-MTX)化疗的最适时间。方法建立骨肉瘤裸鼠动物模型,应用HD-MTX腹腔化疗,在化疗后不同时间应用CF解救10次,观察动物体上肿瘤的坏死率及各器官的毒副作用。结果所有实验动物均出现不同程度的食欲减退和尿量减少,应用HD-MTX化疗后12h开始给CF解救,肿瘤坏死率高,不良反应相对较小;24h后给CF其不良反应明显增高;6h前给CF,虽然不良反应明显小,但其疗效也明显降低。结论HD-MTX化疗后12h开始应用CF解毒疗效好,毒副反应小,安全可行。
Objective To investigate the optimal time of CF treatment in naked mice with osteosatcoma treated with high - dose MTX. Methotis A animal tumor model was developed by cultured MG63 cells. After chemotherapy with HD - MTX, CF was given ten times at different time to alleviate adverse reaction. Pathological examination was performed to observe turmor necrosis rates and toxic and side effects in different organs, Results All animals had gastrointestinal side effects. When CF rescue therapy was given twelve hours after the completion of chemotherapy, turmor necrosis rates were higher and the side effects were relatively less; when CF was given after 24 hours, CF side effects significantly increased; when CF was given before 6 hours, the CF was less effective. Conclusion Twelve hours after the completion of chemotherapy is the optimal time for CF rescue therapy with higher efficacy and less toxicity and side effects.
