摘要
根据SSR分子标记的可转移性和可通用性,利用棉花中已知的SSR引物,筛选适用于海滨锦葵的SSR引物。在筛选的50对引物中,只有1对引物在海滨锦葵中无扩增值点,扩增成功率达到98%。在5株表型差异明显的海滨锦葵中扩增出多态位点的引物有22对,占扩增引物的44.9%。22对产生多态性的SSR引物共扩增出151个等住基因,其中差异等位基因数为87个(占总等位基因数的57.62%);每对引物产生的等位基因数在3-21个(平均数为6.86)、差异等位基因数在1-16个(平均数为3.86)。开发出的适用于海滨锦葵的22对SSR引物可为海滨锦葵的混合交配系统检测和遗传多样性分析提供研究基础和技术支持。
According to the transferability and currency of SSR markers, SSR markers of Kosteletzkya virginica were screened with the reported SSR markers of cotton. Only one of 50 selected SSR primers could not generate bands and the amplified success rate was 98%. 22(44.9% ) of 50 selected SSR primers can generate polymorphic bands among five K. virginica individuals that had obviously phenotypic difference. 22 SSR primers generated polymorphic bands. Total 151 alleles were generated, 87 (57.62%) of which were polymorphic alleles. The number of alleles generated by each primer was from 3 to 21, with the mean of 6.86. 22 SSR primers of K. virgin/ca developed in this study may be provided as reference basis and technological support for the detection of mass breeding system and analyses of genetic diversity.
