摘要
目的:建立测定人血浆中美他环素质量浓度的高效液相色谱法。方法:以土霉素为内标,乙腈-0.05mol·L^-1磷酸二氧钾(30:70)为流动相,流速1.0mL·min^-1,柱温30℃,检测波长244nm,进样量20μL。结果:在0.134~5.373mg·L^-1浓度范围内,线性方程为Y=0.008765+0.068608C,r=0.9992。平均回收率为98.7%,日内RSD〈3,日间RSD〈25。结论:该法具有简单快速、灵敏特异、准确,适用于美他环素的血药质量浓度测定及药动学研究。
OBJECTIVE To establish an HPLC method for the determination of metacycline in human plasma. METHODS Oxytetracycline was used as the internal standard, and detected at 244 nm. The mobile phase consisted of acetonitrile-0. 05 mol·L^-1 potassium dihydrogen phosphate solution (30:70), flow rate was 1.0 mL·min^-1 ,the column temperature was 30℃ ,the injection volume was 20μL. RESULTS The calibration cures was linear in the range from 0. 134μg·mL^-1 to 5. 373μg·mL^-1 for metacycline (r = 0. 999 2). The relative standard deviations of the intra-day and inter-day were less than 5%. The mean ex traction recovery was 98%. CONCLUSION The method is simple, rapid, sensitive and accurate, which may be suitable for the determination of metacycline concentration in human plasma and pharmacokinetics studies.
