摘要
聚合酶链反应扩增的乙酰胆碱受体单克隆抗体重链可变区基因经EcoR1和BamH1双酶切割后,与载体质粒pBluescript连接,并经转化大肠杆菌DH5α而扩增,扩增后的重组质粒经酶切证实含有乙酰胆碱受体抗体的基因。为核苷酸序列分析准备了材料。
The heavy chain variable region gene of a monoclonal antibody (McAb) against acetylcholine receptor (AChR) amplified by polymerase chain reaction (PCR) was transformed into competent E coli DH5α after digested with Eco R1 and Bam H1 and ligated onto plasmid pBluescript.The recombinant plasmid DNA were isolated from E coli DH5α and digested with Eco R1 and Bam H1 again. The results show that the recombinant plasmid DNA contain right size of insert which is the amplified PCR products of the heavy chain variable region of the anti-AChR McAb.
