Sequence analysis of N gene fragment of wild measles virus in Ningbo City.

摘要 目的探讨宁波市麻疹流行株的基因型别和遗传特征。方法利用细胞培养从宁波市2004,2005年28例临床诊断麻疹病例的咽拭子中分离麻疹病毒,通过RT-PCR扩增出病毒核蛋白(N)基因C末端456bp片段进行测序,并分析其和GenBank中麻疹病毒各基因型代表株的同源性。结果分离到8株麻疹病毒,其N基因片段与H1型代表株Hu nan.CHN的同源性为97.6%-98.5%。与麻苗Shanghai-191的同源性为92.8%-93.4%。结论宁波市麻疹流行株属于H1基因型。 Objective To explore the genetic characteristics and genotype of measles virus circulating in Ningbo City. Methods Measles virus was isolated from throat swabs of 28 clinical diagnosed measles patients in 2004 and 2005 and RT - PCR method was employed to amplify N gene fragment of measles virus for sequencing. Then, the homogeneity of the sequence between these measles virus and the representative strains with each genotype in GeneBank were analyzed . Results Eight strains of measles viruses were isolated from specimens. Compared with H1 and the vaccine strain, the homogeneity of nucleotide sequences of N gene fragment was 97.6% - 98.5 % and the homogeneity of nucleotide sequences of N gene fragment was, as compared with that of Shanghai- 191, 92.8 % -93.4%. Conclusion The genotype of measles virus circulation in Ningbo City belongs to HI genotype.
