摘要
用体外液体培养法,观察组胺H_2受体激动剂4-MN对人类早幼粒白血病细胞系HL_(60)细胞的增殖和分化的影响。结果表明,10 ̄(-3)~10 ̄(-5)mol·L ̄(-1)的4-MH能抑制HL_(60)细胞增殖,促进其分化。用胞浆RNA斑点杂交技术检测到10 ̄(-4)mol·L ̄(-4)4-MH能降低HL_(60)细胞c-myc基因的mRNA表达。
he effects of 4-MH(4-methylhistamine,an agonist of histamine H2-receptor)on the prolif-eration and differentiation of the HL60 promyelocytic leukemia cell line were investigated in theliquid cuIture in vitro.It showed that 4-MH at concentrations ranging from 10-5mol·L-1 to 10-3mol·L-1 significantly inhibited the proliferation and induced the differentiation of HL60 cells(P<0.05),The dot blot technique of total cellular RNA with α-32 P nicktranslated c-myc gene asprobe was used to study the changes in transcription level of c-myc oncogene as HL60 leukemiccells were treated bv 4-MH.The results showed that 4-MH 10-4 mol·L-1 could decrease level ofc-myc mRNA and inhibit the transcription of c-myc mRNA in HL60 cells.
