摘要
为了全面、系统、深入地研究有毒中药关木通,采用乙腈-酸水系统,用反相高效液相色谱法分析关木通及其配伍中药中马兜铃酸A的含量,与文献方法相比,缩短了测定时间,并从总体上降低了分析成本。色谱条件是:C18柱,流速:1mL/min。检测波长:250nm。流动相:乙腈:(水+冰醋酸)=58:42。该法精密度高,稳定性好,线性范围为0.07~0.42μg,回归方程为Y=5.0×10^6X-32827,相关系数r=0.9999,加样回收率为99.12%。实验结果表明,关木通与生地、当归、大黄、生甘草等中药配伍后,AAⅠ含量均有不同程度的降低,其中以关木通配伍生地组和大黄组最为明显,分别降低了57%和56%。
To study the chemical toxic components in caulis aristolochiae manshuriensis more deeply, a new reversed phase high performance liquid chromatography(RP-HPLC) method was set up to determine it's content of aristolochic acid A. Compared with literature means, this method shortened the determining time greatly and decreased analyzing cost in the long run. The chromatographic conditions were described as Symmetry ODS C18 column, with a mixture of acetonitrile-acetic acid and water as mobile phase. The flow rate was 1 mL/min and the detective wavelength was 250 nm. The linear range was 0.07 - 0.42 μg (r = 0.9999). The regression equation was Y = 5.0×10^6X - 32827. The average recovery of aristolochic acid A was 99.1%. The results shows that after combined with rchmannia dried root, licorice root, chinese angelica root and rhubarb, the content of aristolochic acid A in caulis aristolochiac manshuriensis decreased with different degree, among them rehmannia dried root and rhubarb arc distinctive, which decreased 57% and 56% respectively. In conclusion, this method is specific, sensitive and with a good repeatability, which is suitable for the determination of aristolochic acid A in caulis aristolochiae manshuriensis.
