摘要
目的通过对三种肺癌患者手术切除标本测定其细胞内信号传递因子(Stat3、Bcl-2)、增殖细胞核抗原(PCNA)的表达水平,以期为肺癌的早期诊断提供新指标。方法采用免疫组织化学方法检测18例鳞癌、21例腺癌,4例小细胞肺癌组织及癌旁组织、正常组织中的Stat3、Bcl^2、PCNA表达水平。结果PCNA在腺癌组织中的阳性表达率为100%,高于癌旁组织(35%)和正常组织(20%)且P均<0.05;Stat3在腺癌组织中的阳性表达率为85%,高于癌旁组织(40%)和正常组织(20%)且P均<0.05;Stat3在腺癌组织上的阳性率(85%)高于bcl-2(25%),且P<0.05。PCNA在鳞癌组织中的阳性表达率为95%,高于癌旁组织(25%)和正常组织(15%)且P均<0.05;Stat3在鳞癌组织上的阳性率(55%)高于bcl-2(30%),且P<0.05。PCNA和Stat3在小细胞未分化癌组织中的阳性表达率均为100%,均高于正常组织(0%~25%)且P均<0.05;PCNA在所有癌组织中的阳性率(97.73%)高于Stat3(72.73%)、bcl-2(34.09%)且P均<0.05。PCNA在无淋巴结转移的癌组织中的阳性率(95.24%)高于Stat361.90%),且P<0.05;Stat3在有淋巴结转移的癌组织中的阳性率(82.61%)高于bcl-2(26.09%),且P<0.05。PCNA和Stat3在癌组织中的阳性细胞数分别为41.068±22.137、37.636±31.465均高于bcl-2的8.590±15.750,且P均<0.05。Stat3在肺癌中的表达与bcl-2和PCNA呈正相关(r分别为0.332,0.472,P均<0.05)。结论PCNA和Stat3可作为肺癌的标志物,且比bcl-2敏感;PCNA作为无淋巴结转移、高分化肺癌的肿瘤标志物均优于Stat3。Stat3可作为判断肺癌尤其是腺癌的指标,且对中分化肺癌或有淋巴结转移肺癌的判断均优于bcl-2。Stat3在肺癌中的表达与bcl-2和PCNA呈正相关。
Objective To analyze the relationship of Star3 expression with tissue types, clinical, stages, age, gender, Bcl-2 and proliferation cell nuclear antigen(PCNA) in human lung carcinoma, and to evaluate the role of Star3 in the pathogenesis and the rumor markers of lung carcinoma. Methods Immunohistochemical method were used to detected Star3, Bcl-2, and PCNA in 18 SC, 21 adenocarcinoma and 4 NSCL specimens, their near tissue and the normal tissue. Results The positive rate of PCNA(100%) in adenocarcinoma was higher than the rate in tissue near cancer(35%), normal tissue (20%) (P〈0. 05). The positive rate of Star3 (85%) in adenocarcinoma was higher than the rate in tissue near cancer(40 %), normal tissue(20 %) (P〈0. 05). The positive rat e of Stat3 (95 % ) in adenocarcinoma was higher than the rate of Bcl-2 in SC tissue(P〈0.05). The positive cells of PCNA and Stat3 in lung carcinoma were more than that Bcl-2(41. 068±22. 137 and 37. 636±31. 465 to 8. 590± 15. 750, both P〈0. 05). A positive correlation of the expression of Stat3 with that of PCNA(r= 0. 472, P=0. 001) and Bcl- 2 (r= 0. 332, P= 0. 027) was identified. Conclusion PCNA and Stat3 could be markers of lung carcinoma. The expression of Star3 in hmg carcinoma with that of PCNA and Bcl-2 was identified.
