摘要
目的探讨荧光定量PCR技术检测大肠癌患者外周血CK19mRNA、CK20mRNA和CEAmRNA的临床意义。方法采用荧光定量PCR技术,检测大肠癌患者76例,健康志愿者30例外周血CK19mRNA、CK20mRNA和CEAmRNA水平。结果76例大肠癌患者外周血CK19mRNA的水平为5 962.5±19 926.1 copies/ml,其中38例阳性,阳性率为50.0%;CK20mRNA的水平为6 244.3±15 113.1 copies/ml,其中28例阳性,阳性率为36.8%;CEAmRNA的水平为7 377.0±26 281.5 copies/ml,其中36例阳性,阳性率为47.4%。对照组30例未见表达。单项检测阳性率较低,联合其中二项检测阳性率较高,联合三项阳性率可达68.4%。结论外周血CK19mRNA、CK20mRNA和CEAmRNA可作为大肠癌微转移辅助诊断指标,联合二至三项指标有较高的检出率,定量检测有助于大肠癌微转移的监测。
Objective To measure CK19mRNA,CK20mRNA and CEAmRNA in peripheral blood of colorectal cancer by Realtime RT-PCR, consequently discussing those results clinical significance. Methods 76 colorectal cancer and 30 volunteers of CK19mRNA,CK20mRNA and CEAmRNA in peripheral blood were detected by Real-time RT-PCR.Results The levels of 76 colorectal cancer CK19mRNA in peripheral blood reach （5 962.5 ＋ 19 926.1） copies/ml, of which 38 patients results axe positive; The levels of 76 rectacancer,coloncancer patients CK20mRNA in peripheral blood reach （6 244.3 ＋ 15 113.1 ） copies/M, of which 28 patients results are positive; The levels of 76 colorectal cancer CEAmRNA in peripheral blood reach （7 377.0 ＋ 26 281.5） copies/ml,of which 36 patients results are positive;contrarily all of those volunteers were not detected CK19mRNA,CK20mRNA and CEAmRNA in their peripheral blood. Conclusion CK19mRNA, CK20mRNA and CEAmRNA in peripheral blood can be as auxiliary index for diagnosing coloreetal cancer, also can monitor the transmission of coloreetal cancer by quantity it.
