摘要
目的建立一种反相高效液相色谱梯度洗脱分离及测定地塞米松磷酸钠、曲安西龙、氢化可的松、曲安奈德、醋酸泼尼松龙、醋酸地塞米松和哈西奈德的方法。方法采用Hypersil ODSC18柱(250mm×4.6mm,5μm),流动相A为甲醇,B为0.2%磷酸二氢钾,C为水,D为乙腈,梯度条件为0→5min,A:33%→38%,B:42%→39%,C:10%→8%,5→20min,A:38%→58%,B:39%→0,C:8%→27%,流速1.0mL·min^-1,柱温30℃,检测波长240nm。结果在20min内分离检测7种激素,7种组分在一定线性范围r均〉0.9993,平均回收率在97.0%~103.0%。结论本法简便快速、灵敏准确、重现性好、方法可行。
OBJECTIVE To propose a gradient elution method for the determination ot 7 adrenocortical hormones by reversed phase high performance liquid chromatography (RP-HPLC). METHODS Adrenocortical hormones separated on a Hypersil ODSC18 column (250 mm×4.6mm, 5μm ) were used. The mobile phase was A: menthanl, 33%→38%; B: 0.2% potassium dihydrogen phosphate, 42%→39%; C: water, 10%→8%; D: acetontrile in 5 min, then A: 38%→58%; B: 39%→0; C: 8%→27% in 5 to 20 min. The flow rate was 1.0 mL · min^-1. The detective wavelength was 240 nm. RESULTS The method showed a good linear relationship between the concentrations and the peak area. The recovery rates were between 97.0% and 103.0%. CONCLUSIONS The method is accurate, convenient and suitable for the determination of 7 adrenocortical hormones.
