冷暴露大鼠心肌线粒体H^+运转ATP酶活力及能量偶联的改变 被引量:8

Changes of H - Transporting ATPase Activity and Function of Energy Coupling of Myocardial Mitochondria in Rats During Cold Exposure

摘要 作者以无机磷释放量的测定比较大鼠心肌线粒体H运转ATP酶水解活力的变化;用ANS荧光强度的变化反映大鼠心肌线粒体内膜的能量偶联功能。发现:(1)线粒体H运转ATP酶水解活力对照组为9.94±0.05Piμmol·min/mg,冷暴露1,2,3,4,5周分别为4.21±0.05,5.02±0.06,2.68±0.04,3.26±0.05,4.71±0.08 Piμmol·min/mg。(2)荧光强度对照组为53.00±2.18相对单位,冷暴露1,2,3,4,5周分别为41.44±2.01,48.00±1.22,28.67±1.32,34.33±2.29,32.78±2.82相对单位。结果表明:冷暴露后的大鼠心肌线粒体H^+运转ATP酶水解活力降低,线粒体内膜的氧化磷酸化偶联功能亦降低。 The activity of submitochondrial H+ -transporting ATPase was assayed by relating of phosphorus. The function of energy coupling was determined by the changes of fluorescence intensity of ANS. It was found that the H - transporting ATPase activity of mitochondria from the rats of the control was 9.94 ± 0.05 Piμmol· min 1/mg the H +-transporting ATPase activity(Pi μmol· min 1/mg) of mitochondria from the rats exposed to the cold for 1, 2, 3, 4 and 5 weeks was4.21±0.05,5.02±0.06,2.68±0.04,3.26±0.05 and 4.71±0.08 respectively. The fluorescence intensity of ANS from the rats of the control was 53.00 ± 218, which is a relative unit; the fluorescence intensities (relative unit ) of ANS from the rats exposed to the cold lor 1, 2, 3, 4 and 5 weeks were 41.44±2.01, 48.00±1.22,28.67 ± 1.32,34.33±2.29 and 32.78±2.82 respectively. It is shown that the activities of H - transporting ATPasc were decreased in the rat heart after cold exposure. The functions of energy coupling arc much lower than that of the control.
