摘要
目的:观察异鼠李素对YTMLC-90、C6、A549、Hela、SMMC-7721和lewis等6种肿瘤细胞株的体外抑制作用。方法:采用不同时间、不同浓度处理靶细胞系,以MTT法检测异鼠李素对肿瘤细胞的生长抑制作用。结果:小剂量梯度(80、40、20、10、5、2.5μg/ml),给药48h和72h对各肿瘤细胞株抑制作用不明显;大剂量梯度(100、80、60、40、20μg/ml),给药72h,Lewis细胞株有明显病变,并随剂量而显示出梯度,IC50为72.18μg/ml;给药96h,各细胞系均有病变,IC50分别为:87.27μg/ml(SMMC-7721),96.69μg/ml(A549),106.23μg/ml(Hela),79.31μg/ml(YTMLC-90),65.01μg/ml(C6),Lewis细胞株抑制显著,无法观察到梯度,未测定OD值。IC50均高于丝裂霉素(P<0.05)。结论:异鼠李素对鼠源性肿瘤细胞Lewis和C6两株细胞抑制作用明显,但体外作用较丝裂霉素弱,如需开发为抗肿瘤药物需作结构修饰。
Objectives To investigate the inhibitory effect of Isorhmnetin on tumor ceil line YTMLC-90, C6, A549, Hela, SMMC-7721 and Lewis. Methods We dealt with target ceils at different times and concentrations. MTI" assay was performed to test the inhibitory effect of isorhmnetin on 6 tumor cell lines. Results Distinct inhibitory effects were not observed for low dosage groups, neither in 48h nor 72h exposure. For high dosage groups, the inhibitory effects were very distinct, especially on Lewis cells. And there were grads with dosages. The IC50 for SMMC-7721, A549, Hela , YTMLC-90 and C6 were 7.27μg/ml, 96.69μg/ml, 106.23μg/ml, 79.31μg/ml, 65. 01μg/ml, respectively. For the inhibitory effect on Lewis was obvious, the grads could not be observed. The OD value was not measured. All IC50 were higher than those of Mitomycin P 〈 0.05 ). Conclusions As an antitumor drug candidate, Isorhmnetin has distinct inhibitory effect on Lewis and C6 ceils, although the effect is lower than Mitomycin. Prospective research is warranted.
