摘要
目的:观察正常豚鼠耳蜗螺旋神经节钙调素的分布,因为作为真核细胞中最重要的Ca2+受体钙调素(CaM)在听觉径路上的分布和功能尚未见报道.方法:用健康白色豚鼠分成实验组和对照组,耳蜗脱钙后行石蜡包埋切片,采用免疫组化ABC法定位螺旋神经节中的CaM免疫反应(CaM-IR).结果:在耳蜗顶至底回螺旋神经节细胞中见CaM-IR分布但形态不一,或淡染或深染,或胞浆内或核及胞浆内均有.结论:说明了耳蜗螺旋神经节细胞内含CaM,并且细胞之间的功能状态可能不同,其CaM可能参与听觉信号的传导以及中枢对耳蜗的调抑功能,这种“上传下达”是通过CaM参与神经递质的合成与释放、轴浆运输完成的.
Objective: Distribution of calmodulin (CaM) in the spiral ganglion (SG) of normal guinea pig cochlea was observed. CaM is the most important Ca 2+ receptor of eukaryotic cells, but little is known about its distribution and function in the auditory passage. Methods: White healthy guinea pigs were divided into the experimental and control groups. After losing the calcium, the cochlea was covered up by paraffin wax and cut into slices. The distribution of CaM in the SG was investigated with a sensitive ABC immunostaining method. Results: There were CaM immunoreaction (CaM IR) in SG cells (SGC) of normal guinea pigs from apical to basal turns. But the positive reaction was different from cells to cells, which were lightly or heavily stained, and in which, cytoplasm or/and nuclei were positive. Conclusion: These results may suggests that CaM occurs in abundance in SGC of guinea pig, those differences may result from different functional status of SGC. CaM may take part in the auditory transmission, which is achieved by the axplasmic transport, the synthesis and release neurotransmitter.
