摘要
制备了5种不同的类风湿关节炎吸附剂.用它们与类风湿关节炎病人血清作静态吸附实验,EL ISA检测吸附前后血清中RF s的浓度变化.实验发现,以纤维素球为载体,环氧氯丙烷活化后固定苯丙氨酸为配基的吸附剂综合吸附效果最好.该吸附剂对病人血清中IgGRF,IgM RF,IgARF的吸附中分别为39.3%,56.0%,28.1%.血液相容性实验结果显示,该吸附剂的血液相容性良好,能够满足临床应用要求.本文还研究了手臂对吸附剂吸附性能的影响,静态吸附实验结果显示PEG 4000为手臂的吸附剂吸附性能要高于以1,4-丁二醇二缩水甘油醚为手臂的吸附剂.
Five different adsorbents i. e. cellulose beads as carrier, Phe or calf thymus DNA as ligand for the removal of rheumatoid factors (RFs) were prepared. Static adsorption experiments were conducted with the serum of RA patient, and the changes in concentration of Rheumatoid factors (RFs) were evaluated by ELISA. The results showed that the adsorbent with phenylalanine directly linked to the cellulose beads via epichlordrin had the highest adsorption capability than all the others. The concentration of IgGRF before adsorption was 524.6 IU/mL, after adsorption that was 318. 4 IU/mL, and the degree of removal was 39.3 % The concentration of IgMRF before adsorption was 805.6 IU/mL, after adsorption was 354.3 IU/mL, the degree of removal was 56.0 %; The concentration of IgARF before ad- sorption was 198.5 IU/mL, after adsorption was 142.8 IU/mL, the degree of removal was 28.1%. From the above results of investigation and other experiments, it is considered that hydrophobicity is important in the reaction between the RFs and Phe. Blood compatibility properties was studied on animal (rabbit) model, the results showed that no significant decrease of blood components occured. Preliminary studies on spacers showed that the PEG4000 spacer has a higher adsorption capability compared to the 1,4-butandioldiglycidylether spacer.
