摘要
从临床表现为繁殖障碍的猪群中分离到一株猪繁殖与呼吸综合征病毒(PRRSV)FJ04A株,用RT-PCR方法进行鉴定,扩增出该分离毒的结构蛋白基因并进行了序列测定.预测其推导的结构蛋白相对分子质量、等电点、抗原位点和糖基化位点等.序列比较结果表明:该分离毒ORFs2-7与美洲型代表株VR2332核苷酸同源性为98.1%-99.5%,氨基酸同源性为96.5%-99.2%;而与欧洲代表株LV的核苷酸同源性为57.3%-63.4%,氨基酸同源性为54.7%-77.6%.表明该分离毒为PRRSV美洲型毒株.
Swine herds in Fujian Province have experienced recently outbreaks of porcine reproductive and respiratory syndrome (PRRS) characterized by abortion and high mortality in pregnant sows. A porcine reproductive and respiratory syndrome virus (PRRSV) was isolated from these herds, which was denominated FJ04A. The structure protein genes of PRRSV FJ04A strain were amplified and sequenced. Molecular analysis of the predicted encoding product of ORFs 2 to 7, such as molecular weight, isoelectric point, epitope and glycosylation site was performed by using several softwares. Sequence analysis revealed that the PRRSV strain shared98.1% -99.5% nucleotide and 96.5% -99.2% amino acid sequence identities to North American genotype VR2332, 57.3% -63.4% nucleotide and 54.7% -77.6% amino acid sequence identities with the Dutch Lelystad strain. The results showed that the PRRSV FJ04A strain belongs to North American genotype.
