摘要
目的:探讨以C_(18)快速色谱柱替代常规色谱柱实现HPLC快速梯度分析时,梯度洗脱程序的调整规律。方法:采用Allti- ma C_(18)高速色谱柱(33mm×7mm,3μm;53mm×7mm,3μm)、Alltima C_(18)常规色谱柱(250mm×4.6mm,5μm)及YMC C_(18)常规色谱柱(150mm×4.6mm,5μm);梯度洗脱;检测波长254nm;流速为0.8~2.5mL·min^(-1)。比较不同梯度洗脱程序下利奈唑胺及其14个有关物质的色谱行为。结果:色谱柱确定后,梯度变化率(ΔC_B/Δt_G)是决定色谱系统选择性的关键因素;在利用快速色谱柱替代普通色谱柱进行分离时,应根据具体分离情况调节流速,使色谱系统达到最佳;当HPLC梯度洗脱系统的流速改变后,应根据洗脱体积(FΔt_G)不变的原则重新确定梯度运行时间。结论:采用高速色谱柱时并相应地调整梯度程序,可顺利实现HPLC梯度洗脱快速分析。
Objective:The adjustment regulation of the HPLC gradient elution method were discussed when high - speed columns instead of general columns. Methods :The analysis was carried on the high -speed columns( Alhima C18 ,33 mm× 7 mm,3μm ;53 mm × 7 μm,3 μm) and the general columns( YMC C18 column, 150 mm×4. 6 mm,5 μm; Alhima C18 column,250 mm × 4. 6 mm,5 μm)with mobile phases in gradient programs. The detection wavelength was 254 nm. The flow rate was 0. 8 - 2. 5 mL· min^-1. The chromatography behaviors of Linezolid and its 14 related substances were compared. Results:When the columns were decided ,the key factor of the selectivity of the chro- matography system was the gradient changing ratio( △CB/△tG). High -speed columns instead of general columns,the mobile phase flow rate should be adjusted according to the separating situation so as to achieve the best chromatography system. When the mobile phase flow rate of the HPLC gradient elution was changed ,the eluting time should be readjusted based on the principle that the eluting volume dose not change( FAtG ). Conclusion :Using high -speed col-umns and modified gradient elution procedures,the high- speed HPLC analysis should be achieved.
