摘要
目的:探索胰岛素样生长因子-1(IGF1)及其受体(IGF1R)在甲状腺乳头状癌发生、发展中的作用,寻找新的甲状腺乳头状癌诊断标志物,并为甲状腺乳头状癌的病因研究寻找新的理论依据。方法:以15例正常人甲状腺组织标本作对照,对62例经手术切除并由病理证实的甲状腺乳头状癌患者的癌、癌旁及正常区组织标本进行研究,应用RT-PCR方法,检测IGF1/IGF1R在甲状腺癌组织与正常人甲状腺组织中的表达。结果:甲状腺乳头状癌组IGF1/IGF1RmRNA的表达量明显高于正常对照组(P<0.01)。甲状腺乳头状癌患者的癌、癌旁及正常区组织中IGF1/IGF1RmRNA的表达量按正常、癌旁、癌组织顺序逐渐增高(P<0.01)。结论:IGF1/IGF1R的高表达促进了甲状腺上皮细胞的异常增殖,提示IGF1/IGF1R可作为甲状腺乳头状癌诊断的标志物。
Objective: To study the role of insulin-like growth factorl and insulin-like growth factorl receptor (IGF1/IGF1R) in the carcinogenesis of thyroid papillary carcinoma. Methods: The mRNA levels of IGF1/IGF1R were assessed by RT- PCR method in thyroid cancer tissues, adjacent tissues and tumor free tissues of 62 patients with thyroid papillary carcinoma and in normal thyroid tissues of 15 healthy controls. Tissue specimens were obtained by biop- sy, and confirmed by histological evaluation. Results: The mRNA levels of IGF1/IGF1R increased in thyroid carcinoma tissues compared with normal tissues of healthy controls ( P 〈 0.01 ). There was a significant difference between thyroid cancer tissues and the tumor free tissues ( P 〈 0.01 ). The mRNA levels of IGF1/IGF1R increased from the tumor free tissues, adjacent tissues to the thyroid cancer tissues ( P 〈 0. 01 ). Conclusions: The high expression of IGF1/IGF1R acts greatly in the carcinogenesis and development of thyroid papillary carcinoma, and they might be used as indicators for early diagnosis of thyroid carcinoma.
