虎芪活血煎对肺纤维化大鼠病理变化和TGF-β_1表达的影响 被引量:8

The Effect of Huqihuoxue Decoct on TGF-β_1 and Pathological Change in Pulmonary Fibrosis Rats Model

摘要 目的:观察中药虎芪活血煎对大鼠肺纤维化的干预作用,并探讨其可能的作用机制。方法:通过气管内注射博莱霉素A5(BLMA5)5mg·kg-1复制大鼠肺纤维化模型,给予小、中、大剂量(分别为10g·kg-1·d-1、20g·kg-1·d-1和40g·kg-1·d-1)的虎芪活血煎干预后,用HE及VG染色观察肺组织病理变化和胶原沉积情况,并计算肺泡炎和肺纤维化积分;用免疫组织化学(IHC)染色测定肺组织转化生长因子-β1(TGF-β1)表达水平,并行图像定量分析。结果:模型对照组肺泡炎和肺纤维化积分及肺组织TGF-β1表达显著高于正常对照组(P<0.01);虎芪活血煎中、大剂量组肺泡炎和纤维化积分及肺组织TGF-β1表达均显著低于模型对照组(P<0.01);而虎芪活血煎小剂量组作用不佳。结论:中、大剂量的虎芪活血煎能减轻肺泡炎和肺纤维化程度,对博莱霉素A5诱导的大鼠肺纤维化有良好的保护作用。其机制可能是通过抑制TGF-β1的表达,减少了胶原沉积,使炎症及纤维化病变减轻。 Objective: To evaluate the effect and mechanism of Huqihuoxue Decoction on bleomycin-induced pulmonary fibrosis in rats. Methods: Firstly, to reproduce rat models of pulmonary fibrosis by injecting bleomycin As (5 mg/kg) into trachea, then to treat the rat models by use of Huqihuoxue Decoction with low - , median - and high-dose (10g·kg^-1·d^-1、20g·kg^-1·d^-140g·kg^-1·d^-1 respectively), finally, the pulmonary histopathologieal changes and collagen disposition were observed by hematoxylin and eosin (HE) staining and Van Gieson (VG) staining, and the expression of transforming growth factor- 1 (TGF - β1 ) in lungs were measured by immunohistochemieal (IHC) staining. Results: The scores of the alveolitis and pulmonary fibrosis, and the expression of TGF- β1 in lungs were lower in Huqihuoxue Decoction with median - and high-dose group than those in model group ( P 〈 0.01), while those in Huqihuoxue Decoction with low-dose group comparing with model group were shown no significant difference. Conclusion: Huqihuoxue Decoction with median- and high-dose would ameliorate the alveolitis and pulmonary fibrosis from bleomytin-induced pulmonary fibrosis in rats. Inhibiting the expression of TGF - β1, decreasing collagen deposition in pulmonary tissues may be one of the mechanisms.
