哺乳动物细胞的生存要求适当的氧化与抗氧化平衡,而活性氧中间产物则破坏这一平衡且在细胞凋亡中起重要作用。氧化应激介导的细胞凋亡,可能与活性氧中间产物对DNA损伤导致的聚ADP核糖转移酶活化和P53的积累以及脂质过氧化物使细胞内Ca2+水平增加有关。
Mammalian cells exist in a state of oxidative siege in which survival requires an appropriate balance of oxidants and antioxidants. Reactive oxygen intermediates (ROI) break this balance and play an important role in apoptosis. Oxidative stress mediated DNA damage has been shown to elicit the activation of poly ADP ribose transferase and the accumulation of P53 and lipid hydroperoxides result in marked increase in cellular Ca 2+ , all of which are associated with apoptosis. ROI may also mediate apoptosis through the activation of an oxidative stress responsive nuclear transcription factor such as NF кB.
