摘要
实验选用雄性Sprague-Dawley大鼠研究顺铂肾毒性发生的机理。实验动物静脉注射10mg/kg顺铂后3小时,肾脏光镜与透射电镜检查及肾小球滤过率无明显变化。但顺铂注射后30分钟,尿钠、钾浓度分别增加56%及2.6倍,3小时后肾小管细胞线粒体功能明显异常:状态4呼吸上升1倍,状态3呼吸、呼吸控制率及FCCP解偶联呼吸分别抑制46%、74%及47%。揭示线粒体功能障碍可能为大白鼠顺铂肾毒性发生早期的中心环节。
Male Sprague-Dawley rats received an intravenous cisplatin of 10 mg/kg. Three hours following cisplatin administration,no evident morphological abnormalities were found by both light and electron microscopy; there was also no significant changes in glomerular filtration rate, However, 30 minutes after cisplatin injection, urine sodium and potassium concentration increased by 56% and 260% of those in control animals,respectively.Apparent renal mitochondrial respiration dysfunction was encountered in cisplatin-treatd rats 3 hours later: state 4 respiration was doubled and state 3 respiration, respiratory control ration and carbonyl cyanide p-trifluoromethoxyphenylhydrazone-uncoupled respiration decreased by 46%、74% and 47% of those in controls, respectively. The present data suggest that mitochondrial dysfunction may be a very early event in cisplatin nephrotoxicity in rats.
