IL-6和TNF-α在大鼠脑水肿发生中的动态变化及意义 被引量:3

摘要 目的:探讨白细胞介素-6(IL-6)和肿瘤坏死因-α(TNF-α)在脂多糖(LPS)致大鼠脑水肿发生中的含量变化及其作用。方法:用LPS诱发大鼠脑水肿发生后,制备脑组织匀浆,以ELISA法测定IL-6含量,放免法测定TNF-α含量。结果:LPS注入后,实验大鼠脑组织中TNF-α、IL-6释放增加,与空白对照组和生理盐水组比较,有极显著性差异(P<0.01)。LPS组脑组织含水量与Evans blue(EB)含量,TNF-α与脑组织含水量,IL-6与脑组织含水量,IL-6与EB含量均呈正相关,r值分别为0.537、0.423、0.473、0.831,P<0.05。结论:IL-6、TNF-α介导脂多糖致脑水肿的发生、发展过程,与脑组织炎性损伤密切相关。 Objective :To study the roles of IL-6 and TNF-α in the onset of brain edema induced by lipopolysaccharide in rat. Methods:RIA and ELISA methods were used to measure the contents of TNF-α and IL-6 in brain tissue in vivo after brain edema induced by LPS in rat. Results:The levels of TNF-α and IL-6 in brain tissue in LPS group were increased compared with A group and NS group(P〈0. 01 ). In LPS group,water content in brain tissue and Evans blue (EB) content,TNF-α and water content in brain tissue ,IL-6 and water content in brain tissue ,IL-6 and EB content all had positive correlation (r=0. 537, 0. 42a, 0. 437, 0. 831 in turn, P〈0.05). Conclusion: IL-6 and TNF-α participate in the onset and development of brain edema induced by LPS and have close relationship with inflammatory brain injury.
