摘要
目的观察缺血及缺血再灌注(IR)大鼠肾脏诱导型一氧化氮合酶(iNOS)表达变化的规律。方法制作大鼠肾脏缺血及IR模型,不同时间摘取肾脏,免疫组织化学法检测iNOS表达变化。HE染色观察肾脏损伤程度。结果缺血时及IR后iNOS表达均明显增强(P<0.05),且随时间呈一定的变化规律,呈先升高后下降趋势,24 h达峰值,72 h时降至2 h水平。结论肾脏缺血早期iNOS即起损伤作用,并于再灌注后加重损伤。
Objective To investigate the expression levels and the roles of induced nitric oxide synthase(iNOS) in rats' kidney after interruption of blood flow and the following ischemia - reperfusion(IR) injury. Methods Rats were divided into 3 groups: interruptions (5,15,30 min), IR(2,5,8,24,48,72 h) ,control groups. They were killed at different time points. Kidney tissue samples were studied by method. Results iNOS increased significantly( P 〈 0.05) in ischemia and IR groups. The expressions of iNOS were significantly higher than those of control group( P 〈 0.05). It changed with a certain regulation according to the course. Conclusion The results suggest that iNOS has some injury effect during the early stage of ischemia and the effect becomes worse during the following IR.
