胆红素吸附树脂的筛选及安全性检测 被引量:2

摘要 目的从多种合成树脂中筛选出胆红素吸附率高且安全性好者以清除腹水中的胆红素。方法10种待测吸附树脂,1#、2#为阳离子交换树脂,功能基团为-NH2;3#、4#为大孔吸附树脂(孔径20~50nm);5#为NK-107树脂,7#为NK-110树脂;6#为大孔吸附树脂(孔径30~50nm,极性,聚苯乙烯);8#为阳离子交换树脂(丙烯酸),功能基团为-COOH;9#为大孔强碱性苯乙烯阴离子交换树脂,功能基团为-N+(CH3)3;8#、9#树脂按1∶1比例混合为10#。树脂筛选按1∶10比例加入腹水(体积比),计算吸附率,实验重复6次。安全性测定:①用紫外分光光度计扫描树脂浸液上清,测定溶出物;②以家兔升温法检测热原。结果3#、5#、7#、8#、9#、10#树脂的DBiL与IBiL吸附率均较高。以紫外分光光度计检测3#、5#、7#、8#、9#、10#6种树脂浸液的上清,结果显示基本不含有溶出物;经家兔升温法行树脂热原测定,3#树脂不符合测定规定,其余树脂均符合测定规定。结论5#、7#、8#、9#、10#树脂吸附率较高且安全,可进一步试用于临床。 Objective To screen out the appropriate adsorbents with good safty and higher bilirubin adsorption rate from a number of synthetic resins for removal of ascitic bilirubin. Methods All of 10 adsorption resins, 1^#, 2^# were positive ion charring resin,with function group of -NH2; 3^#, 4^# were large-pore adsorption resin (aperture 20~50 nm); 5^# was NK-107 resin, 7^# was NK-110 resin; 6^# was large-pore adsorption resin (aperture 30~50 nm, polarity, polystyrene); 8^# was positive ion charring resin (acrylic acid), with function group of -COOH; 9^# was large-pore adsoption resin of strong alkali phenylethene, with function group of -N^+(CH3)3; 10^# was admixture resin with 8^# : 9^# = 1 : 1. The different resins were screened with resin/ascites of patient at the ratio of 1 : 10. The experiments were repeated 6 times, then the adsorption rates were calculated. The safety test was performed as follows: ① The supernatent of resin extracts was screened by ultraviolet light spectrophotometer. Its dissolved matters were determind. ② The pyrogen was detected by rising temperature technique in rabbits. Results The adsorption rates for both indirect bilirubin and direct bilirubin in 3^#, 5^#, 7^#, 8^#, 9^#, 10^# resin were considerably high. No dissolved matters of resins were found, only in 3^# resin the test for pyrogen was positive. Conclusion The experiments of 5^#, 7^#, 8^#, 9^#, 10^# resin showed higher bilirubin adsorption rate and safety. Thus they can be used in further clinical study.
