摘要
目的探讨干预协同刺激信号对反复自然流产患者Th1/Th2转换的影响。方法用人绒毛膜癌细胞株JEG-3制备滋养细胞抗原;用该抗原刺激反复自然流产患者外周血单个核细胞,并分为两组:实验组加入细胞毒性T细胞相关抗导4(CTLA4Ig,10μg/mL组和1μg/mL组),对照组加IgG(10μg/mL组和1μg/mL组),培养72h,分别取24h和72h上清液,采用酶联免疫吸附法(ELISA)测上清液中白介素2(IL-2)、干扰素γ(IFN-γ)和白介素4(IL-4)的水平。结果与同质量浓度的对照组相比,CTLA4Ig组产生的IL-4水平明显高于对照组(P<0.05),而产生的IL-2水平明显低于对照组(P<0.05),IFN-γ水平两组间差异无统计学意义(P>0.05)。与CTLA4Ig1μg/mL组相比,CTLA4Ig10μg/mL组产生的IL-4水平明显升高(P<0.05),IL-2水平明显降低(P<0.05),IFN-γ水平两组间差异无统计学意义(P>0.05)。结论CTLA4Ig能通过阻断CD80/CD86共刺激信号,使反复自然流产患者Th1/Th2细胞因子向Th2型细胞因子偏移。
Objective To assess the influence of blockade of costimulation on Th1/Th2 Cytokines shift in unexplained early recurrent spontaneous abortion (URSA). Methods The levels of IL-2, IFN-γ and IL-4 were determined by ELISA in the supernatant of cultured maternal peripheral blood mononuclear cells(PBMC) from 30 patients of URSA. Cytokine synthesis was induced by activation with antigens from a chorlocarcinomer cell line of JEG-3, with CTLA4Ig at concentrations of 10 μg/mL and 1 μg/mL (experiment groups), or with IgG at the same concentrations (control groups). Results The CTLA4Ig groups showed significantly higher levels of Th2 cytokines IL-4 (P〈0. 05), lower levels of Thl cytokines IL-2 (P〈0. 05) and the same levels of IFN-γ(P〉 0. 05), compared with the control groups of the same concentrations respectively. The same results were found between the experiment groups of different concentration. Conclusion CTLA4Ig could switch T cell responses from Thl to Th2 in URSA by blockade of CD80/CD86 costimulation.
