摘要
目的:探讨强骨胶囊对雌激素受体(ER)活性的影响。方法:将含有ERs配体结合域(LBD)的质粒pCMV-BD和pFR-Luci报告基因及内标质粒pFRTlaczeo共转染到Hela细胞中,检测强骨胶囊与雌激素受体的关系;MTT法检测强骨胶囊对细胞活力的影响。结果:低剂量(12.5ng/ml)的强骨胶囊对两种ER均无明显效果,但是高剂量(125ng/ml和1.25μg/ml)可以显著地激活ERs,并可以显著提高细胞活性。结论:强骨胶囊治疗骨质疏松的疗效可能与激活ERs的作用有关。
Objective: To study the effects of "Qianggu Capsule" on estrogen receptors activity. Methods: pCMV-BD plasmid containing ERs ligand binding domain (LBD) and pFR-Luci report plasmid and pFRTlaczeo control plasmid were co-transfection into Hela cells, the relationship between "Qianggu Capsule" and estrogen receptors was detected by luciferase activity; the effects of "Qianggu Capsule" on cell activity were detected by MTI" method. Results: Low dose(12.5 ng/ml)"Qianggu Capsule" had no significant effect on ERs, high-dose(125 ng/ml and 1.25 μg/ml) "Qianggu Capsule" obviously activated ERs and improved cell activity. Conclusion: The treatment of osteoporosis by "Qianggu Capsule" may be related to its effect of activating ERs.
