均匀设计优化人血白蛋白荧光猝灭法测定水中Cu(Ⅱ) 被引量:2

摘要 目的:建立了HSA荧光猝灭法测定Cu(Ⅱ)的分析新方法。方法:在均匀设计优化的反应条件下,研究了Cu(Ⅱ)对HSA的荧光猝灭效应。结果:在pH=8.0的KH2PO4-Na2HPO4缓冲溶液中,一定量的Cu(Ⅱ)对HSA荧光的猝灭有良好的线性关系。检出限为19.7ng/ml;相对标准偏差0.9%-1.3%;样品加标回收率93.0%~102.7%;线性范围0.07~6.00μg/ml。结论:方法与单因素轮换法实验结果比较,实验条件更优化,方法更简便、灵敏。可直接用于环境水样中Cu(Ⅱ)的测定。 Objective:To develop a new method for determination of Copper( Ⅱ ) in environmental water with human serum albumin(HSA). Methods:The method is based on the fluorescence quenching effect of Copper(Ⅱ) to HSA in conditions, which were determinated by uniform design experiment, such as KH2 PO4 - Na2 HPO4 buffer solution was pH = 8. 0, reaction temperature was 5℃ and volumn of 1.0 mol/L NaCI solution was 1.0 ml. Results: △I (the quenching value of Ⅰ350 nm) was linear to the concentration of Copper( Ⅱ ) in the range of 0. 07 - 6.0 μg/ml in conditions selected(λex = 290 nm, λem = 350 nm et al. ). The detection limit was 19. 7 ng/ml. The RSD was 0. 9% - 1.3% and the recovery was 93.0% - 102. 7%. Conclusion:These results showed much better than those of Single - factor rotates experiment. The method proposed has been successfully applied to the direct determination of Copper( Ⅱ) in environmental water.
